چکیده

chromatin-associated DNA-directed DNA polymerase activity, and the soluble RNase-sensitive endogenous DNA polymerase activity were examined in two fibroblast cell lines (WI-38 and WI-38VA13), in thirteen lymphoid cell lines originated from normal persons or from acute lymphocytic leukemia, acute myelocytic leukemia, Burkitt lymphoma, or multiple myeloma patients, and also in cells obtained directly from patients with acute lymphocytic leukemia, acute myelocytic leukemia, multiple myeloma, or lymphosarcoma. RNase-sensitive endog enous DNA polymerase activity, which was 1/1000 to 1/10 of DNA-directed soluble DNA polymerase activity, was lowest in Wl-38 cells and highest in lymphoblasts from acute lymphocytic leukemia patients. The soluble and chromatinassociated DNA-directed DNA polymerase activity of WI38V13 cells, compared to that of WI-38 cells, and the activity of acute myelocytic leukemia, Burkitt lymphoma, and multiple myeloma cells, compared to that of normal cells, were two to ten times higher; in contrast, the activity of cells of acute lymphocytic leukemia or lymphosarcoma origin was one-third to one-half that of normal cells.

DETECTION AND RESTRICTION ANALYSIS OF C YTOMEGALOVIRUS DNA PERSISTING IN HUMAN ATHEROSCLEROTIC PLAQUES USING POLYMERASE CHAIN REACTION

The polymerase chain reaction (PCR) as applied to detection of a foreign DNA in clinical specimens could provide a sensitive instrument for rapid detection of viral DNA persisting in tissues of patients suspected of latent infection. Human cytomegalovirus (HCMV) DNA was found in arterial plaques of patients with atherosclerotic lesions using a PCR assay with nested primer oligonucleotides ...

Stimulated normal human lymphocytes contain a ribonuclease-sensitive DNA polymerase distinct from viral RNA-directed DNA polymerase.

Ribonuclease-sensitive DNA synthesis is demonstrated in a cytoplasmic particulate fraction of normal human blood lymphocytes stimulated with phytohemagglutinin, but not in unstimulated lymphocytes. DNA polymerase purified from this fraction does not transcribe the heteropolymeric regions of 70S RNA from RNA tumor viruses, thus distinguishing this enzyme from the RNA-directed DNA polymerase (rev...

Relationship between RNA-directed DNA polymerase (reverse transcriptase) from human acute leukemic blood cells and primate type-C viruses.

An RNA-directed DNA polymerase was isolated from the peripheral blood leukocytes of a patient with acute myelomonocytic leukemia by successive purification of a particulate cytoplasmic fraction with endogenous, ribonuclease-sensitive DNA polymerase activity. Like RNA-directed DNA polymerase from mammalian type-C virus, the human leukemic cell enzyme efficiently utilized (A)(n).(dT)(12-18) and (...

RNA-dependent DNA polymerase of an endogenous type C virus of mice: purification and partial characterization.

An RNA-dependent DNA polymerase was isolated from purified virions of endogenous oncornaviruses released by the MOPC-315 murine myeloma cell line. Upon sodium dodecyl sulfate-polyacrylamide gel electrophoresis, the purified enzyme was found to consist of two major polypeptides with molecular weights of about 28,000 and 26,500. The active enzyme had a molecular weight of approximately 56,000, as...

Genetic evidence for two protein domains and a potential new activity in bacteriophage T4 DNA polymerase.

Intragenic complementation was detected within the bacteriophage T4 DNA polymerase gene. Complementation was observed between specific amino (N)-terminal, temperature-sensitive (ts) mutator mutants and more carboxy (C)-terminal mutants lacking DNA polymerase polymerizing functions. Protein sequences surrounding N-terminal mutation sites are similar to sequences found in Escherichia coli ribonuc...
